What HPLC does

A chromatogram plots detector response against retention time. Peaks represent signals detected under the method; they are not automatically a complete inventory of every substance in a sample.
How area percentage is calculated
A reported area percentage usually compares the integrated area of a selected peak with the total integrated area included in the calculation. The result is method-dependent: detector wavelength, integration settings, sample preparation, gradient, column, and which peaks are included can all matter.
Because of that, a high area percentage should be read as a chromatographic result under stated conditions. It is not a synonym for potency, sterility, clinical effectiveness, or regulatory approval.
Retention time is useful, but not enough
Retention time can help compare a sample peak with a reference or an established method. It is affected by column chemistry, mobile phase, temperature, flow rate, and instrument conditions. Matching one retention time does not by itself prove complete identity.

What to inspect on a chromatogram
- Method name, column, detector, wavelength, and run conditions
- Retention time of the principal peak
- Integration boundaries and whether minor peaks are visible
- Reported area percentage and calculation description
- Sample or lot identifier and test date
- Whether a complementary identity method is reported
HPLC does not answer every quality question
HPLC purity is one analytical attribute. It does not answer whether a material is sterile, endotoxin-free, correctly labeled, stable in every condition, or suitable for human or veterinary use. Those questions require separate evidence and, in some cases, a regulated manufacturing and clinical framework.

Common questions
Is 99% HPLC purity the same as 99% clinical effectiveness?
No. HPLC area percentage is an analytical result under a stated method and does not measure clinical effectiveness.
Does HPLC prove identity?
HPLC can support characterization, but identity is stronger when supported by appropriate complementary methods and clear sample traceability.
